Microbial ATP Kit HS is intended for determination of intracellular ATP in bacterial cells. ATP Reagent SL especially developed for the assay of mithocondrial ATP metabolism. The reagent has a decay of ATP and light emission of less than 1% per minute. The stable light allows manual handling and you can use a simpler luminometer. The detection limit is 10^-15 mole ATP, making the reagent suitable for assays of mithocondrial ATP production rate down to the detection limit and for other continous monitoring of enzymatic reactions where the magnesium ion concentration has to be adjusted.
Reagent HS is a High Sensitivity (HS) reagent with high levels of luciferase which results in a moderate ATP- and light emission decay (-6%/min). The high activity also has the effect that any contamination caused during reconstitution is quickly degraded. The reagent is suitable for measurements down to 10^-17 mole ATP and is stable enough to use with a luminometer with or without dispensers. Please note that 12-071 ATP Reagent HS is in a size especially developed for hygiene control purposes.
A crucial step in the ATP assay is the extraction of the ATP from the cells, when the cells are opened and the ATPases in the cell neutralized. The process has to be very fast, since if the ATPases are not neutralized immediately they will start degrading the ATP.
If the extractant is not neutralized after use it will degrade the luciferase in the ATP reagents when added. This is avoided by reconstituting the ATP Reagents in Diluent B as the diluent contains components to neutralize the extractant.
